ABSTRACT

Objective: To assess the effects of Manuka honey, Ocimum sanctum, Curcuma longa, and 0.2% chlorhexidine mouthwash on Streptococcus mutans and Lactobacillus acidophilus levels. Material and Methods: A randomized controlled trial will be conducted on dental students of Teerthanker Mahaveer Dental College and Research Centre, Moradabad. The study participants will be divided into four groups. Each group will have a total of 20 individuals. By using a lottery system, Group A (Manuka honey mouthrinse), Group B (Ocimum sanctum mouthrinse), Group C (Curcuma longa mouthrinse), and Group D (0.2% chlorhexidine mouthrinse) will be chosen. To match the circadian cycle, saliva will be collected at baseline and again after 2 weeks between 10 and 10.30 a.m. The sterile container will subsequently be delivered to the microbiological laboratory and processed as soon as possible to measure Streptococcus mutans and Lactobacillus acidophilus count. For 2 weeks, participants were told to use 10 mL of mouthrinse twice daily. Results: The mean oral hygiene index?simplified (OHI?S) score of all the four groups showed reduction in their scores from baseline to after the study period. For both S. mutans and L. acidophilus, there was a substantial Percentage Reduction (PR) between the prerinse and postrinse samples in all four groups. Discussion: Because quantitative actions play a crucial part in the caries disease process, the changes in microbial activity before and after administration of experimental mouthwashes were examined. Conclusion: Essential oil aqueous extracts were as efficient antibacterial mouthwashes as chlorhexidine and iodine mouthwashes

ABSTRACT

Objective:To investigate the efficacy of compound Lactobacillus acidophilus tablets combined with bismuth-containing quadruple therapy in the treatment of Helicobacter pylori-positive (Hp +) chronic non-atrophic gastritis and its effect on Lactobacillus and Escherichia coli.Methods:A total of 104 patients with Hp + chronic non-atrophic gastritis who received treatment in Affiliated Hospital of Shaoxing University from March 2019 to September 2020 were included in this study. They were randomly divided into control and observation groups, with 52 patients in each group. The control group was treated with bismuth-containing quadruple therapy. The observation group was treated with compound Lactobacillus acidophilus tablets (2 tablets once, three times per day) based on bismuth-containing quadruple therapy. Two groups were treated for 2 successive weeks. The endoscopic grading score of gastric mucosa, improvement in pathological grading of gastric mucosa, Hp eradication rate, and Lactobacillus and Escherichia coli levels were compared between the two groups. Results:After treatment, endoscopic grading score of gastric mucosa (gastric mucosa inflammation, inflammation activity, atrophy, and intestinal metaplasia) in the observation group was (1.31 ± 0.18) points, (1.24 ± 0.18) points, (1.15 ± 0.16) points, (1.04 ± 0.15) points, respectively, which was superior to (1.73 ± 0.25) points, (1.59 ± 0.21) points, (1.44 ± 0.19) points, (1.30 ± 0.18) points in the control group ( t = 3.67, 3.80, 4.17, 4.52, all P < 0.01). Improvement in pathological grading of gastric mucosa in the observation group was significantly higher than that in the control group [94.23% (49/52) vs. 78.85% (41/52), χ2 = 4.04, P < 0.05]. Hp eradication rate in the observation group was significantly higher than that in the control group [92.31% (48/52) vs. 75.00% (39/52), χ2 = 4.50, P < 0.05]. After treatment, the amount of Lactobacillus and Escherichia coli in the observation group were (8.30 ± 0.95) copies and (8.04 ± 0.95) copies, which were significantly greater than (7.51 ± 0.93) copies and (8.30 ± 0.95) copies in the control group ( t = 4.02, 3.75, both P < 0.01). Conclusion:Compound Lactobacillus acidophilus tablets combined with bismuth-containing quadruple therapy is highly effective on Hp + chronic non-atrophic gastritis and it can effectively improve intestinal microflora.

O objetivo geral do presente estudo foi avaliar a aplicação dos jatos de plasma de baixa temperatura sob pressão atmosférica (PBTPA) produzidos por gás de argônio e hélio como gases de trabalho, no controle de biofilmes cariogênicos. Para tanto, foram estabelecidos os parâmetros físicos dos PBTPA gerados com argônio e hélio que se mostraram efetivos frente a biofilmes mono, dual e polimicrobianos compostos por combinações das espécies Streptococcus mutans, Streptococcus gordonii, Streptococcus sanguinis, Lactobacillus casei, Lactobacillus acidophilus, Candida albicans e Actinomyces naeslundii. Os biofilmes mono, dual e multi-espécies foram submetidos ao tratamento com PBTPA produzidos por dois dispositivos diferentes, um obtido comercialmente (kINPen09®) que usou argônio como gás de trabalho, e outro protótipo desenvolvido pela FEG-UNESP (Faculdade de Engenharia de Guaratinguetá) que usou hélio. Análises quantitativas e microscópicas (confocal, microscopia eletrônica de varredura) foram realizadas. Foi incluído controle negativo (sem tratamento), positivo (clorexidina 0,12%) e controle de gás, utilizando apenas fluxo de gás, sem produzir plasma. Além disso, os efeitos celulares do PBTPAargônio e hélio sobre biofilme dual e multi-espécies também foram analisados em microscopia eletrônica de varredura e microscopia de varredura a laser confocal. Todos os ensaios foram realizados em triplicata em três experimentos independentes. Os resultados foram tabulados e analisados quanto à distribuição. A seguir, os testes estatísticos mais adequados foram selecionados. O nível de significância foi de 5%. Os resultados obtidos para os tratamentos dos biofilmes mono, dual ou multi-espécies com PBTPA-argônio e hélio foram todos significativos em comparação ao controle negativo em todos os tempos analisados. Para PBTPA-argônio, não houve recuperação de S. gordonii e S. sanguinis em todos tempos analisados. Para PBTPA-hélio, os melhores resultados foram obtidos em 5 e 7 minutos de exposição dos biofilmes ao PBTPA. Finalmente, tanto o dispositivo gerador de PBTPA que trabalhou com gás argônio quanto o dispositivo que trabalhou com gás hélio, demonstraram resultados promissores e poderão contribuir para o desenvolvimento de novos protocolos de Odontologia de Intervenção Mínima. (AU) The general objective of this study was to evaluate the application of lowtemperature plasma under atmospheric pressure (PBTPA) of argon and helium flow, in the control of cariogenic biofilms. For this, the effective physical parameters of PBTPA-argon and helium in mono, dual and polymicrobial biofilms composed of combinations of the species Streptococcus mutans, Streptococcus gordonii, Streptococcus sanguinis, Lactobacillus casei, Lactobacillus acidophilus, Candida albicans and Actinomyces naeslundii were established. The multi-species biofilms were treated by different PBTPA generating devices, one obtained commercially (kINPen09®) that used argon as working gas, and another prototype developed by FEG-UNESP (Faculdade de Engenharia de Guaratinguetá) that used helium as working gas. Quantitative and microscopic analyzes (confocal, scanning electron microscopy) were performed. Negative control (no treatment), positive control (chlorhexidine 2%) and gas control (argon) were included. Besides that, cellular effects of PBTPA-argon and helium on dual and multi-species biofilms were analyzed by scanning electron microscopy (SEM) and confocal laser scanning microscopy. The results obtained for the treatments of mono, dual or multispecies biofilms with both PBTPA-argon and helium were all significant when compared to the negative control at all times analyzed. For PBTPA-argon, there was no recovery of S. gordonii and S. sanguinis at all analyzed times. For PBTPA-helium, the best results were obtained at 5 and 7 min of exposure of biofilms to PBTPA. All the tests were carried out in triplicate in three independent experiments. The results are tabulated and analyzed in terms of distribution. Next, the most suitable statistical tests were selected. The level of significance was 5%. The results obtained for the treatments of mono, dual or multi-species biofilms with PBTPA-argon and helium were all significant compared to the negative control at all analyzed times. Finally, both PBTPA generating could contribute to the development of new protocols for Minimal Intervention Dentistry (AU)

Background: Caries is a multifactorial disease, microbes being an important etiological factor. “Probiotics” are nonpathogenic microbes which can stimulate health-promoting flora and suppress pathogens. It's the era of prevention, clinical approach toward prevention by altering bacterial ecology is the need of the hour. The aim of the study was to evaluate anticariogenic effect of probiotic curd (Amul's Probiotic Dahi) containing Lactobacillus acidophilus (La5) and Bifidobacterium lactis (Bb12) in 6–12 years of age group. Materials and Methods: This was a prospective, randomized, nonblinded study conducted on school children aged between 6 and 12 years having Decayed, Missing, and Filled Teeth and/or deft ?2. Selected subjects were divided into two groups of 31 each, study group received probiotic curd (“Amul's Probiotic Dahi”) two times a day in a quantity of 100 g, respectively, for 21 days and control group did not receive any intervention. Salivary Streptococcus mutans count and salivary pH estimation were done at the baseline, after 7 days and at the end after 24 days, for comparison. Results: No statistical difference was seen in salivary S. mutans count between both groups, after intervention of 7 days. However, there was significant difference in salivary S. mutans count at the end of 24 days. There was no statistical difference in salivary pH between both groups. Conclusion: The present study concludes that probiotics substitute decreases salivary S. mutans count after continuous consumption for 3 weeks and there is no short-term effect on salivary pH.

Introducción: El uso de los aceites esenciales de plantas medicinales ha demostrado poseer propiedades antibacterianas frente a diversas especies bacterianas. Objetivo: Identificar la composición química del aceite esencial de Minthostachys mollis (Griseb) L. y determinar su actividad antibacteriana frente a Streptococcus mutans y Lactobacillus acidophilus. Métodos: Se realizó un estudio experimental in vitro. Se empleó el software EpiInfoTM para el cálculo de las repeticiones. Se obtuvo aceite esencial de M. mollis proveniente de la región altoandina del Perú. La composición química fue analizada por cromatografía de gases acoplada a espectrometría de masas. La actividad antibacteriana se evaluó por medio del método de difusión en disco, además, se identificó la concentración mínima inhibitoria y la concentración mínima bactericida. Resultados: Los principales constituyentes del aceite esencial fueron mentona (32,9 por ciento) y eucaliptol (28,06 por ciento). El aceite esencial fue efectivo para inhibir el crecimiento de S. mutans y L. acidophilus, con halos de inhibición de 19,040 ± 0,883 mm y 18,008 ± 0,861 mm. La clorhexidina al 0,12 por ciento fue más efectiva que el aceite esencial para inhibir el crecimiento de L. acidophilus (p < 0,05). Una concentración de 1,6 por ciento (v/v) del aceite esencial fue inhibitoria y bactericida frente a S. mutans; para L. acidophilus una concentración de 3,2 por ciento (v/v) fue inhibitoria. Conclusiones: Los principales constituyentes del aceite esencial de M. mollis fueron mentona y eucaliptol y demostró ser efectivo para inhibir el crecimiento de S. mutans y L. acidophilus(AU)


Introduction: The use of essential oils from medicinal plants has been shown to possess antibacterial properties against various bacterial species. Objective: To identify the chemical composition of the essential oil of Minthostachys mollis (Griseb) L. and determine its antibacterial activity against Streptococcus mutans and Lactobacillus acidophilus. Methods: An experimental study was performed in vitro. EpiInfoTM software was used to calculate the repetitions. Essential oil of M. mollis was obtained from the high Andean region of Peru. The chemical composition was analyzed by gas chromatography coupled to mass spectrometry; antibacterial activity was evaluated by means of the disk diffusion method, in addition the minimum inhibitory concentration and the minimum bactericidal concentration were identified. Results: The main constituents of the essential oil were menthone (32.9 percent) and eucalyptol (28.06 percent). The essential oil was effective in inhibiting the growth of S. mutans and L. acidophilus, with inhibition halos of 19.040 ± 0.883 mm and 18.008 ± 0.861 mm. Chlorhexidine 0.12 percent was more effective than essential oil in inhibiting the growth of L. acidophilus (p < 0.05). A concentration of 1.6 percent (v/v) of the essential oil was inhibitory and bactericidal against S. mutans; for L. acidophilus a concentration of 3.2 percent (v/v) was inhibitory. ABSTRACT@#Lactobacillus acidophilus (L. acidophilus) is one of the etiological agents for dental caries and dominant in the deep carious lesion. L. acidophilus has also been identified in persistent root canal infection and also related to the failure of endodontic treatment. Photodynamic therapy is a therapeutic process involving the combination of a nontoxic photosensitizer and a light source. The excited photosensitizer reacts with reactive oxygen species (ROS), which induce injury and death of the microorganism. This study aimed to prove the effect of irradiation time of photodynamic therapy to the number of L. acidophilus. Forty-two Eppendorf tubes were treated with 0.5 ml L. acidophilus distributed into seven groups. Group 1 as the control group received no treatment. Groups 2, 3, 4, 5, 6 and 7 were treated with a combination of 0.5 ml toluidine blue O (TBO) as a photosensitizer and 630 nm photoactivated (Fotosan®) exposure time for 10, 20, 30, 40, 50 and 60 sec. Then, all were stored in an incubator of 37ºC for 48 h. Later, the colony-forming unit (CFU) was counted for each group. There were significant differences in the number of L. acidophilus in CFU of the various irradiation times. The longer the photodynamic therapy irradiation was, the lesser the number of live L. acidophilus became. At 50 sec and 60 sec irradiation, none of the L. acidophilus was found alive.

ABSTRACT Objective: To investigate the antimicrobial activity of colloidal selenium nanoparticles in chitosan solution (Cts-Se-NPs) against Streptococcus mutans, Lactobacillus acidophilus, and Candida albicans. Material and Methods: Cts-Se-NPs solution was prepared using a simple chemical reduction method. The MIC and MBC against S. mutans, L. acidophilus, and C. albicans were determined using the broth dilution assay. Results: The Cts-Se-NPs had remarkable antimicrobial activity against S. mutans, L. acidophilus, and C. albicans. The MIC values of the Cts-Se-NPs were lowest for S. mutans (0.068 mg/ml) compared to L. acidophilus (0.137 mg/ml), and C. albicans (0.274 mg/ml). The MBC values of the Cts-Se-NPs against the microorganisms after one, two, six, and 24 hours indicated that the concentration of 0.274 mg/ml of Cts-Se-NPs completely killed S. mutans, L. acidophilus, and C. albicans after one, two, and six hours, respectively. At the concentration of 0.137 mg/ml, S. mutans and L. acidophilus were killed after six and 24 hours, respectively. Conclusion: These findings encourage the potential use of Cts-Se-NPs in dentistry, while further clinical research is required in this area.

Abstract In neonates, rapid change in diet imbalances gut health allowing colonization of opportunistic pathogens that confer harmful effects on animal health causing reduced digestion and malabsorption of nutrients. In this milieu, probiotic feeding can be a promising approach in promoting animal health and stabilization of gastrointestinal microbiota. Hence, the present study was designed to investigate the effect of Lactobacillus acidophilus NCDC15 enriched fermented milk on nutrient digestibility, faecal biomarkers and immune response in Murrah buffalo calves. Twenty-four, neonatal calves (5-7 days) were randomly allocated into four groups for 90 days. The control group (CT) was provided a basal diet of calf starter and green fodder (maize and jowar), without any probiotic fermented milk (PFM) supplementation. Basal diet was supplemented with probiotic fermented milk at 100, 200 and 300 mL/calf/day, in PFM100, PFM200 and PFM300 groups, respectively. Nutrient digestibility remained similar among the groups. Faecal acetate was higher (P<0.05) in PFM200 and PFM300, while, faecal butyrate was increased (P<0.05) with all levels of probiotic supplementation than control. Faecal Lactobacillus and Bifidobacterium count were increased (P<0.05) with a concomitant reduction in coliform population (P<0.05) among all the treatments. Cell-mediated and humoral immune response were higher (P<0.001) in PFM200 and PFM300 than CT. Overall, it can be concluded that inclusion of Lactobacillus acidophilus NCDC15 in the form of fermented milk upto 300 mL/calf/day improved immunity and faecal biomarkers in Murrah buffalo calves without any adverse effect on nutrient utilization which may positively impact growth performance in Murrah buffalo calves.

OBJECTIVE@#To investigate the effects of Da-Cheng-Qi Decoction (DCQD, ) combined with Lactobacillus acidophilus (LA) on the recovery of gastrointestinal (GI) function in traumatic brain-injured (TBI) mice.@*METHODS@#A total of 150 male C57BL/6 mice were randomly divided into sham-injury, normal saline (NS), DCQD (0.4 mL/day), LA (⩾1 × 10 cfu/day LA), DCQD+LA (LA administration at the same dosage after 4 h of feeding DCQD), and ½ DCQD+LA groups (LA administration at the same dosage after 4 h of feeding ½ DCQD dose) by a random number table, 5-8 mice in each group. The sever TBI model was constructed according to Feeney's enhanced gravitational forces of free falling. On days 1, 3, and 7 post-TBI, plasma diamine oxidase (DAO) and D-lactic acid levels were assessed by enzyme-linked immunosorbent assay (ELISA). Occludin expression in the intestinal epithelium was assessed by Western blot analysis. Transmission electron microscopy (TEM) was used to observe the morphological changes in the network structure of interstitial cells of Cajal (ICC) and change of enteric nervous system-ICC-smooth muscle cell (ENS-ICC-SMC). Immunofluorescence staining was used to detect changes in the network structure of the ICC.@*RESULTS@#Compared with the NS group, occludin expression in the DCQD+LA group significantly increased on Day 1, 3, and 7 post-TBI (P<0.05 or P<0.01). The concentration of DAO significantly decreased in the LA, DCQD, and DCQD+LA groups on Day 3 and 7, whilst the D-lactate concentrations in the LA and ½ DCQD+LA groups decreased on Day 1 and 3 post-injury (P<0.05 or P<0.01). The NS group experienced a great damage on the ENS-ICC-SMC network morphology and ICC network structure, and all treatment groups had some improvements, among which the DCQD+LA group presented relatively intact network morphology.@*CONCLUSIONS@#DCQD combined with LA treatment could effectively repair the intestinal mucosal barrier and improve GI motility in mice after TBI. The combination of DCQD and LA was more effective than their respective monotherapies.

ABSTRACT

Background: Wound treatment still a task for medical professionals, according to the time advancement. We need to elaborate further new interventions to cope up this common factor of community. The objective of the study was to assess the anti-inflammatory topical role of Lactobacillus acidophilus with antibiotic in wound repair of rats based on of microscopic parameters. This comparative study was conducted in the Department of Anatomy Al-Tibri Medical Collage and Hospital Isra University Karachi Campus from December 2018 to April 2019.Methods: Total 18 male of wistar rats were randomly selected for this study. The study comprised of three topical groups Control, Antibiotic and Lactobacillus acidophilus groups respectively.Results: The samples were taken from the wound site for the preparation of microscopic slides, to count the numbers of neutrophils, lymphocytes and macrophages for the evaluation of anti-inflammatory process in both groups on day 3 and 7. Data obtained were analyzed by SPSS version 20 by applying One Way ANOVA and post hoc Tukey’s test.Conclusions: The results were shown potent anti-inflammatory effects of Lactobacillus acidophilus group in comparison with other conventional therapy. The study concludes that the topical application of Lactobacillus acidophilus had remarkable anti-inflammatory effects in wound management.

Background: Diarrhoea is the second most common cause of under-five mortality especially in developing world. Many studies have been conducted so far using different probiotic strains with variable outcome. So, the aim of the present study was to compare the clinical efficacy of Bacillus clausii and multi strain probiotic formulation as adjunct treatment of acute diarrhoea.Methods: This prospective single blind randomized controlled clinical trial included 300 infants and children between 6 months to 6 years of age admitted in a tertiary care hospital Sylhet, Bangladesh with acute watery diarrhoea having varied dehydration status ranging from no to severe dehydration excluding shocked state. Cases were randomly assigned to three groups which were group I (n=100) comprised of children who were treated with standard treatment (according to WHO guideline) only as control group, group II (n=100) who received standard treatment plus Bacillus clausii and group III (n=100) who received standard treatment plus multi strain probiotic formulation (Lactobacillus casei, Lactobacillus rhamnosus, Lactobacillus acidophilus, Lactobacillus bulgaricus, Bifidobacterium breve, Bifidobacterium infantis, Streptococcus thermophilus). Primary outcome variables were duration, frequency of diarrhoea and consistency of stool. Secondary outcome variable was duration of hospital stay.Results: Mean duration of diarrhoea was significantly shorter (p=0.001) in group III (2.62 days) compared to group I (3.26 days) and group II (3.22 days). Frequency of diarrhoea was significantly lower on day 3 of probiotic administration in group III (p <0.05) and on day 5 of treatment in group II (p <0.05). Stool consistency significantly improved on day 3 in group III (p <0.05) while it was on day 4 in group II. The duration of diarrhoea, hospital stay, stool consistency and frequency of stool on day 3 were not statistically significant (p >0.05) in group II in comparison to group I and group III.Conclusions: Multistrain probiotic formulation is effective in reducing the duration, frequency of diarrhoea and duration of hospital stay.

ABSTRACT: Technique of complex coacervation was used to produce microcapsules of Lactobacillus acidophilus La-5 encapsulated in gelatin and gum arabic which were then freeze-drying. Microcapsules were characterized using scanning electron and optical microscopy, and resistance of probiotics was evaluated during release into a simulated gastrointestinal tract and storage at different temperatures. The complex coacervation process produced microcapsules with a high encapsulation efficiency (77.60% and 87.53%), ranging from 127.14-227.05 μm with uniform distribution. Microencapsulation was an efficient approach to achieve significant protection of probiotics against simulated gastrointestinal conditions compared with free cells. Encapsulation also improved the viability of probiotics during storage at either −18 ºC for 120 days, 7 ºC for 105 days or 25 ºC for 45 days. Therefore, complex coacervation was demonstrated to be adequate and promising for encapsulation of probiotics.


RESUMO: A técnica de coacervação complexa foi utilizada para a produção de microcápsulas contendo Lactobacillus acidophilus La-5 em gelatina e goma arábica seguida de secagem por liofilização. As microcápsulas foram caracterizadas por microscopia óptica e eletrônica de varredura, assim como a resistência dos probióticos frente à liberação "in vitro" ao trato gastrointestinal simulado e ao armazenamento em diferentes condições de temperatura também foram avaliados. O processo de coacervação complexa formou microcápsulas com alta eficiência de encapsulação (77,60% e 87,53%), tamanho compreendido entre 127,14 e 227,05 µm e distribuição uniforme. As microcápsulas foram eficientes em promover a proteção substancial dos probióticos frente às condições gastrointestinais simuladas, em comparação às células livres. A encapsulação também foi eficiente em manter a viabilidade dos probióticos durante o armazenamento em temperaturas de −18 ºC por 120 dias, 7 ºC por 105 dias e 25 ºC por 45 dias. Dessa forma, a coacervação complexa se mostra adequada e promissora para a encapsulação dos probióticos.

BACKGROUND: Atopic dermatitis (AD) in infants is often related to food allergies (FA). The beneficial effects of lactic acid bacteria towards allergic diseases have been reported, but there are few reports on their effect and preferable dosages on AD in young children with concomitant FA. OBJECTIVE: To examine additional effects of two different dose of paraprobiotic Lactobacillus acidophilus L-92 (L-92) on the clinical treatment in young children afflicted by AD with diagnosed or suspected FA. METHODS: Fifty-nine AD young children from 10 months to 3 years old, with FA or who had not started to ingest specific food(s) because of high specific IgE levels, were recruited and randomly allocated into L-92 group (daily intake of 20 mg L-92/day) and placebo group. Participants were given test sample with conventional treatment for AD over a 24-week period. The severity of eczema was evaluated using SCORing Atopic Dermatitis (SCORAD) index before intervention, and at 4, 12, and 24 weeks after intervention. RESULTS: After 24 weeks of intervention, a significant decrease in SCORAD was observed only in the L-92 group when compared with the baseline values. Significant decreases in thymus and activation-regulated chemokine (TARC) and total IgE were also detected 24 weeks after intake in the L-92 group compared with the placebo group. CONCLUSION: It was suggested that intake of sufficient amounts of L-92 works as an adjunctive treatment of young children afflicted by AD with diagnosed or suspected FA.

ABSTRACT BACKGROUND: Colorectal cancer is one of the major causes of death worldwide. Many studies have been done on the biology of its formation as well as its treatment in recent years. One of the factors involved in the formation or treatment of this malignancy can be attributed to the microbial flora in the intestine. OBJECTIVE: This study investigate the potential preventive effect of Lactobacillus acidophilus and Lactobacillus plantarum in patients with polyps or colorectal cancer (CRC). METHODS: A total of 77 samples were selected in the form of three groups including individuals suffering from CRC, polyps and healthy subjects. Genomic DNA of fecal specimens and standard strains were extracted and amplified employing primers targeting of the 16S rRNA gene for initial detection. Absolute Real Time PCR quantification was used to determine the copy of the bacterial expression per gram of feces. RESULTS: No significant difference were observed between age and gender in the mentioned groups (P=0.06). The average copy number of Lactobacillus acidophilus shows Significant difference between the healthy group and those with polyps (P<0.0001), the healthy group and those with colorectal cancer (P<0.0001), as well as those with polyps and the colorectal cancer patients (P<0.0001). CONCLUSION: These results may indicate that taking Lactobacillus acidophilus in people with a family history of CRC and people with polyps may be a way of preventing, treating or reducing the severity of CRC.


RESUMO CONTEXTO: O câncer colorretal é uma das principais causas de morte em todo o mundo. Muitos estudos têm sido feitos sobre a biologia de sua formação, bem como o seu tratamento nos últimos anos. Um dos fatores envolvidos na formação ou no tratamento desta malignidade pode ser atribuído à flora microbiana no intestino. OBJETIVO: Este estudo investigou o potencial efeito preventivo de Lactobacillus acidophilus e Lactobacillus plantarum em pacientes com pólipos ou câncer colorretal (CCR). MÉTODOS: Um total de 77 amostras foram selecionadas e três grupos foram formados, a saber, indivíduos portadores de CCR, pólipos e indivíduos saudáveis. O DNA genomico de espécimes fecais e de amostras padrão foi extraído e amplificado empregando primers que focalizaram o gene do rRNA 16S para a deteção inicial. A quantificação do PCR em tempo real absoluto foi utilizada para determinar a cópia da expressão bacteriana por grama de fezes. RESULTADOS: Não foram observadas diferenças significativas entre idade e sexo nos grupos citados (P=0,06). O número médio de cópias de Lactobacillus acidophilus mostra diferença significativa entre o grupo saudável e aqueles com pólipos (P<0,0001), o grupo saudável e aqueles com câncer colorretal (P<0,0001), bem como aqueles com pólipos e câncer colorretal pacientes (P<0,0001). CONCLUSÃO: Estes resultados podem indicar que a ingestão de Lactobacillus acidophilus em pessoas com antecedentes familiares de CCR e pessoas com pólipos pode ser uma forma de prevenir, tratar ou reduzir a gravidade da CCR.

Abstract The study of metabolomics requires extracting as many metabolites as possible from a biological sample. This study aimed to determine the optimal method for the extraction of metabolites from solid-state fermented cottonseed meal (FCSM). The UPLC-Q-TOF-MS global metabolomics technology was used to detect the metabolites in FCSM, and the extraction quantity and extraction efficiency of seven different extraction methods, specifically the WA, 50MeOH, 50MeOHB, 50MeCNB, 80MeOHB, 80MeOH and AMF methods were evaluated. The results showed that the number of VIP metabolites extracted by AMF method are 196 and 184 in ESI+ and ESI- mode respectively, it is the largest number of all exacted methods; and the AMF methods also provided a higher extraction efficiency compared with the other methods, especially in indoleacrylic acid, dl-tryptophan and epicatechin (p < 0.01). As a result, AMF/-4 °C method was identified as the best method for the extraction of metabolites from FCSM by Lactobacillus acidophilus. Our study establishes a technical basis for future metabolomics research of fermented feed.

Objetivos: Determinar la eficacia in vitro del efecto inhibidor del extracto etanólico de propóleo en comparación al gluconato de clorhexidina frente a cepas de Lactobacillus acidophilus. Material y Métodos: El tipo de investigación fue experimental, prospectivo, analítico y longitudinal, llevándose a cabo en el laboratorio de microbiología de la Universidad Privada Norbert Wiener durante el año 2016. La población estuvo conformada por cepas de Lactobacillus acidophilus de uso comercial, con una muestra de 30 cultivos en placas petri con agar base sangre, colocándose discos de papel Whatman N° 40 embebidos con extracto etanólico de propóleo al 15% y 30%, así como gluconato de clorhexidina 0,12% como control positivo y alcohol de 70° como control negativo. Se desarrolló el cultivo en condiciones de anaerobiosis a 37 °C. Resultados: Se obtuvo que la concentración al 15% y 30% del extracto etanólico de propóleo presenta un mayor efecto inhibidor promedio de 8,15 mm y 11,75 mm a las 48 horas y de 11,40 mm y 14,25 mm a las 72 horas, respectivamente; en relación al halo de inhibición promedio producido por el gluconato de clorhexidina al 0,12% a las 48 y 72 horas, que fue de 6,55 mm y 8,00 mm. Conclusiones: El extracto etanólico de propóleo al 30% es más efectivo in vitro que la clorhexidina al 0,12% para el control de Lactobacillus acidophilus.


Objectives: To determine the efficacy in vitro of the antibacterial inhibitory effect of the ethanolic extracts of propolis in comparison to the chlorhexidine gluconate against strains of the Lactobacillus acidophilus. Material and methods: The type of investigation was experimental, prospective, analytical and longitudinal. It took place in the laboratory of microbiology at the private university Norbert Wiener during the year 2016. The population consisted of strains of Lactobacillus acidophilus commercial use and the sample consisted of 30 crops of this microorganism in petri plates with blood agar base and where disks embedded paper was placed with ethanol propolis extracts of 15% and 30%, chlorhexidine gluconate of 0.12% as a positive control and alcohol of 70% as a negative control. Underconditions of anaerobiosis at 30°C. Results: The obtained that the concentration at 15% and 30% ethanolicpropolis extract has a higher average effect inhibitor 8.15 mm and 11.75 mmat 48 hours and 11.40 mm and 14.25 mm at 72 hours respectively. In relation to the average inbition halo produced by the chlorhexidine gluconate 0.12% at 48 and 72 hours, which was 6.55 mm and 8.00 mm. Conclusions: The ethanolic extract of propolis30% is more effective in vitro than 0.12% chlorhexidine for the control of Lactobacillus acidophilus.

ABSTRACT

The most common causes of acute viral diarrhea in infants and toddlers are rotavirus, astrovirus and norovirus. The purpose of this study was to evaluate epidemiological data of pathogens obtained from stool exams and compare them with the clinical course in pediatric patients with symptoms of viral acute diarrhea and to investigate the clinical efficacy of Lactobacillus acidophilus (L. acidophilus) for acute diarrhea caused by rotavirus. Clinical data for three types of viral acute diarrhea were compared with the viral detection results by enzyme-linked immunosorbent assay. Viruses were detected in 65.8% of subjects, with rotavirus being the most commonly reported in 63.3% of subjects. To examine the characteristics of each virus, a clinical epidemiological study was performed for 482 cases. Noroviral infection symptoms included vomiting and diarrhea in patients of all age groups. Dehydration in noroviral acute diarrheal patients was less common than in rotaviral acute diarrheal patients. The clinical efficacy of orally administered L. acidophilus in the treatment of acute viral diarrhea in infants and toddlers was also evaluated. L. acidophilus was a probiotic adjuvant in viral acute diarrhea in infants and toddlers.

Resumen En los últimos años se ha reconocido el potencial de las bacterias ácido lácticas (BAL) como agentes bioconservantes de productos cárnicos, incluido el pescado, al reducir el deterioro microbiológico y químico. El objetivo de este trabajo fue determinar el efecto bioconservante de dos cepas de BAL y el tiempo de impregnación en la calidad microbiológica y química de filetes de tilapia. Se evaluaron dos tiempos de impregnación (1h y 2h) y las BAL Lactobacillus plantarum JCM 1149 y Lactobacillus acidophilus ATCC4356. Los filetes bioconservados (FB) se almacenaron a 5°C y se evaluaron los cambios en el recuento de BAL, mesófilos, psicrófilos y coliformes totales durante 30 días. Los recuentos en los FB fueron de 5.94 de Log UFC/g. de BAL, y <2.7 log UFC/g de coliformes y psicrófilos. Los filetes control presentaron recuentos de 1.2 log UFC/g de BAL mientras los coliformes totales y psicrófilos superaron el límite permitido para consumo humano luego de 10 días. Luego de 10 días. El tratamiento que mantuvo la calidad microbiológica y química de los FB por más tiempo fue L. plantarum con inmersión de una hora. Las BAL estudiadas mostraron un efecto inhibitorio de la microflora deteriorante del pescado y una reducción en la formación de nitrógeno volátil, siendo un método viable para la conservación del pescado.


Abstract In recent years, it has recognized the potential of lactic acid bacteria (LAB) as biopreservatives agents for meat products, including fish, by reducing the microbiological and chemical spoilage. The aim of this study was to determine the effect of two strains biopreservative LAB and impregnation time on the microbiological and chemical quality of tilapia fillets. Two impregnation time (1h and 2h) and two LAB (Lactobacillus plantarum JCM 1149 and Lactobacillus acidophilus ATCC4356 were evaluated. The biopreserved fillets (BF) were stored at 5 ° C and the changes of the LAB count, mesophilic, psychrophilic and total coliforms., were evaluated for 30 days. The microbiological analysis show for the BF 5.94 log CFU / g. of LAB and <2.7 log CFU / g of coliform and psychrophilic bacteria. The control Fillets showed 1.2 log CFU / g of LAB while the psychrophile and total coliforms exceeded the allowed limit for human consumption after 10 days. The treatment kept the microbiological and chemical quality of the BF longer was L. plantarum with one hour of impregnation. The LAB studied showed an inhibitory effect of the spoilage microflora of fish and a reduction in the formation of volatile nitrogen compounds, being a viable fish preservation method.

O propósito deste estudo foi determinar o efeito bioconservante de duas cepas de BAL e a avaliação do tempo de impregnação requerido para obter uma boa qua lidade microbiológica e química de filetes de tilapia. Foram avaliados dois tempos de impregnação (1h e 2h) com as BAL Lactobacillus plantarum JMC 1149 e Lactobacillus acidophilus ATCC4356. Os filetes biopreservados (FB) foram estocados a 5oC e valoradas as modificações na contagem de microorganismos BAL, mesófilos, psicrófilos, coliformes totais durante 30 dias. As contagens microbianas nos FB foram de 5.94 de log UFC/g e <2.7 log UFC/g de coliformes e psicrofilos. Os filetes controle apresentaram valores de 1.2 log UFC/g de BAL, não entanto os coliformes totais e psicrófilos superaram o limite permitido para consumo humano após 10 dias de estocagem. O tratamento que manteve a qualidade microbiológica e química dos FB por maior tempo foi de imersão com Lactobacillus plantarum por uma hora. As BAL estudadas amostraram um efeito inibitório da microflora deteriorante dos peixes e uma redução na formação de nitrogênio volátil se apresentan do como um método viável para a preservação de filete de peixe

ABSTRACT

The objective of this work was to adapt a synbiotic aerated diet dessert, produced with the addition of a probiotic culture of Lactobacillus acidophilus La-5 and prebiotic ingredients (fructooligosaccharides and inulin), from the previously developed sucrose-containing formulation, and to evaluate the effects of its ingestion on adult volunteers with metabolic syndrome (MetS) during a period of 8 weeks of intervention. In addition, to improve the resistance of the probiotic to simulated gastrointestinal conditions, a microencapsulation process was optimized. For the development of the product, the formulations were produced in triplicates, in which probiotic culture survival, instrumental texture and sensory acceptability were evaluated up to 112 days of storage under freezing (-18 °C). Subsequently, a randomized, double-blind, placebo-controlled trial was carried out in which the product developed was administered to forty-five volunteers with MetS assigned into two groups, each receiving 40 g/day of: synbiotic diet mousse (SDM) (n=23) and placebo diet mousse (PDM) without pro- and prebiotics (n=22). Fasting blood samples were collected at the beginning and after 8 weeks of daily consumption of both mousses to determine the anthropometric, biochemical, haematological, inflammatory, and immunological parameters. Afterward, with the goal of improving the survival of L. acidophilus La-5 to in vitro simulated gastrointestinal conditions, the microencapsulation process conditions of the probiotic strain via spray drying were optimized using inulin as the encapsulating agent. The viability of L. acidophilus La-5 incorporated into SDM was above 7.8 log CFU/g and remained stable throughout storage. PDM showed lower acceptability (5.77-6.50) after storage than SDM (6.67-7.03). The texture was the most appreciated attribute and hardness of the SDM increased during storage, but remained stable for PDM. The clinical trial revealed significant reductions of total cholesterol and HDL-cholesterol, as well as of immunoglobulins (A and M), and interleukin-1ß in both groups during the intervention period. However, regarding intergroup changes, there were not any significant differences for all parameters evaluated (p>0.05). After the optimization of the microencapsulation process of the probiotic culture (80 mL/min, 82% and 10%, respectively for feed flow, aspiration rate, and inulin concentration), the microencapsulated probiotic strain incorporated in the SDM mousse showed the highest in vitro gastrointestinal survival (p<0.05) in the different stages of the assay, as follows: after the gastric phase: 5.68 log CFU/g (83.3%), the enteral phase I: 5.61 log CFU/g (82.3%), the enteral phase II: 5.56 log CFU/g (81.4%). Therefore, these results suggest that the presence of probiotic and prebiotics in SDM did not provide an additional effect on the health of volunteers with MetS. Additionally, the results confirm the appropriateness of the spray drying process to microencapsulate L. acidophilus La-5 using inulin as coating agent, providing increased resistance to the microencapsulated probiotic strain under in vitro gastrointestinal stress


O objetivo deste trabalho foi adaptar uma sobremesa aerada simbiótica diet do tipo musse, processada com a adição de uma cultura probiótica de Lactobacillus acidophilus La-5 e de ingredientes prebióticos (fruto-oligossacarídeos e inulina), a partir da formulação contendo sacarose desenvolvida anteriormente, e avaliar os efeitos de sua ingestão em voluntários adultos com síndrome metabólica (MetS) durante um período de 8 semanas de intervenção. Adicionalmente, para melhorar a resistência do probiótico frente às condições gastrintestinais simuladas, otimizou-se um processo de microencapsulação da cepa probiótica. Para o desenvolvimento do produto, as formulações foram produzidas em triplicata, em que se avaliou a sobrevivência da cultura probiótica, a textura instrumental e a aceitabilidade sensorial até 112 dias de armazenamento sob congelamento (-18 oC). Em seguida, foi realizado um estudo randomizado, duplo-cego e controlado por placebo, no qual o produto desenvolvido foi administrado a quarenta e cinco indivíduos com MetS divididos em dois grupos, cada um recebendo 40 g/dia de: mousse simbiótica diet (SDM) (n=23) e musse placebo diet (PDM) sem componentes pro- e prebióticos (n=22). As amostras sanguíneas foram coletadas em jejum no início e após 8 semanas de consumo diário de ambas as musses para a determinação dos parâmetros antropométricos, bioquímicos, hematológicos, inflamatórios e imunológicos. Posteriormente, com o intuito de melhorar a sobrevivência do L. acidophilus La-5 em condições gastrointestinais simuladas in vitro, as condições de processo de microencapsulação da cepa probiótica via spray drying foram otimizadas, utilizando inulina como agente encapsulante. A viabilidade de L. acidophilus La-5 incorporados na SDM foi superior a 7,8 log UFC/g e se manteve estável ao longo do armazenamento. A PDM mostrou menor aceitabilidade (5.77-6.50) após o armazenamento do que a SDM (6.67-7.03). A textura foi o atributo mais apreciado, sendo que a dureza da SDM apresentou elevação, enquanto a da PDM manteve-se estável. O ensaio clínico revelou reduções significativas de colesterol total, colesterol-HDL, imunoglobulinas (A e M) e interleucina1ß em ambos os grupos durante o período de intervenção. Entretanto, no que se refere às mudanças intergrupos, não se observou diferenças significativas para todos os parâmetros avaliados (p>0,05). Após a otimização do processo de microencapsulação da cultura probiótica (80 mL/min, 82% e 10%, respectivamente para o fluxo de alimentação, taxa de aspiração e concentração de inulina), a cepa probiótica microencapsulada incorporada a amostra SDM apresentou a maior sobrevivência gastrointestinal in vitro (p<0,05) nas diferentes etapas do ensaio, a saber: após a fase gástrica: 5,68 log UFC/g (83,3%); fase entérica I: 5,61 log UFC/g (82,3%); fase entérica II: 5,56 log UFC/g (81,4%). Portanto, esses resultados sugerem que a presença de probiótico e prebiótico na SDM não apresentou efeitos adicionais na saúde dos voluntários com MetS. Adicionalmente, os resultados confirmaram a adequação do processo de spray drying para a microencapsulação de L. acidophilus La-5 utilizando inulina como agente de revestimento, proporcionando uma maior resistência da cepa probiótica microencapsulada às condições gastrintestinais simuladas in vitro.

Objective To observe the effectiveness of lactobacillus acidophilus combined with electroacupuncture on gastrointestinal function in patients with traumatic brain injury. Methods A total of 167 patients with traumatic brain injury treated in our hospital from June 2013 to December 2016 were randomly divided into combining treatment group (n=84) and lactobacillus acidophilus group (n=83). The lactobacillus acidophilus group was given routine treatment and lactobacillus acidophilus. The combining group was given the routine treatment, electroacupuncture treatment and lactobacillus acidophilus. Data of gastronintestinal function, infection, pH of gastric juice, serum two amine oxidase (DAO) and D-lacticacid level were compared between two groups. Results Compared with lactobacillus acidophilus group, the incidence rates of reflux (χ2=7.948), vomit (χ2=7.514), diarrhea (χ2=4.521), abdominal distension (χ2=5.968), pulmonary infection (χ2=4.521) and enterogenic infection (χ2=4.531) were significantly decreased in the combining treatment group (P<0.05). After 20-day treatment, pH value of gastric juice (t=8.125) was significantly increased, the serum DAO value (t=6.383) and D-lacticacid level (t=6.745) were decreased, in combining treatment group than those of lactobacillus acidophilus group (P<0.05). Conclusion Lactobacillus acidophilus combined with electroacupuncture can improve the gastrointestinal function, stimulate gastric acid secretion and decrease serum levels of DAO and D-lacticacid.
